Specificity

Clone XBR2-401 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Tyrosine-protein kinase transmembrane receptor ROR2. It targets an epitope within the Kringle domain from the extracellular region.

From naïve rabbit antibody library (as described by Peng, H., et al. (2017). J. Mol. Biol. 429(19): 2954 2973).

Application

Quality Control Testing

Evaluated by Immunocytochemistry in T47D cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected ROR2 in T47D cells.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected ROR2 in one million T47D cells.

Western Blotting Analysis: A representative lot detected ROR2 in Western Blotting applications (Peng, H., et. al. (2017). J Mol Biol. 429 (19); 2954-2973).

Affinity Binding Assay: A representative lot of this antibody bound recombinant fragment of Human ROR2 with a KD of 2.5 x 10-6 in an affinity binding assay.

Target description

Tyrosine-protein kinase transmembrane receptor ROR2 (UniProt: Q01974; also known as EC:2.7.10.1, Neurotrophic tyrosine kinase, receptor-related 2) is encoded by the ROR2 (also known as NTRKR2) gene (Gene ID: 4920) in human. ROR2 is a hom*odimeric, single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 34-403), a transmembrane domain (aa 404-424), and a cytoplasmic domain (aa 425-943). Its protein kinase domain is localized in amino acids 473-746. ROR2 is expressed at high levels during embryonic development and its expression levels decrease significantly around day 16 and adults display much reduced levels. ROR2 is involved in the early formation of chondrocytes and may be involved in cartilage and growth plate development. It phosphorylates tyrosine 3-Monooxygenase (YWHAB), leading to induction of osteogenesis and bone formation. It displays very little tyrosine kinase activity in vitro. Higher expression of ROR2 has been reported in several cancers, including solid malignancies, such as renal cell adenocarcinoma, breast cancer, and hematologic malignancies, such as multiple myeloma. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
